About: Steve is
a research technician for Dr. Keith Fowke. He has a 4 year BSc. major in
microbiology from the University of Manitoba. Steve provides tech support and have been
involved in many projects that include many techniques. This includes
molecular biology, cytometry, cell culture, and other protocols. I also run
and overlook our BD FACScailbur cytometer and Epics Elite sorter in level 3. He
has recently begun using Luminex bead technology to study samples.

About:
The majority of new HIV infection occurs via heterosexual transmission. Understanding the factors influencing the susceptibility/protection to HIV infection is important in order to develop a safe and efficient compound to prevent HIV infection. Despite to be at high risk of HIV acquisition, some individuals remain HIV uninfected. This study aims to understanding the immune factor at the female genital tract compartment that might explain this protection against HIV infection.

Impact of Genetic Variation at the CD4 Locus on HIV Susceptibility and Disease Progression

About:
Our lab has previously identified a polymorphism in the CD4 gene that is associated with increased susceptibility to HIV infection and accelerated disease progression. A nearby gene, GNB3, mediates signaling cascades initiated by the HIV co-receptors CCR5 and CXCR4 and contains multiple polymorphisms that regulate its function. The CD4 gene is also in close proximity to LAG-3 (Lymphocyte activation gene 3), a T cell activation and exhaustion marker whose role in HIV infection is not well described. This project will investigate whether the GNB3 825T polymorphism is associated with HIV susceptibility or disease progression among high- and low-risk Kenyan women, and will characterize the expression and function of LAG-3 on immune subsets during HIV infection and ARV therapy.

Investigation of Immune Quiescence: Assessing the role of T regulatory cells a in HIV Exposed Sero-negative (HESN) subjects |
About: Tregs are a subset of CD4+ cells that have important roles in central tolerance and maintenance of homeostasis of immune cells. They have been shown to suppress CD4+ and CD8+ T cells responses to HIV and other antigens. Recently it has been shown that HIV resistant women have higher levels of CD4+CD25+FOXP3+ Tregs compared to HIV-susceptible women. The focus on my project will be to assess the role of Tregs in the Immune Quiescence model of resistance, which suggests that a reduced activation state is protective against HIV infection. To assess if elevated Tregs are contributing to the Immune Quiescent phenotype in HIV resistant women, Tregs will be depleted from PBMCs and analyzed for activation, proliferation and cytokine expression by flow cytometry following stimulation with different activating agents .We hypothesize Treg depletion will result in higher activation, proliferation and cytokine responses, suggesting that Tregs have a suppressive role in HIV resistant women and thus might be contributing to Immune Quiescence. |
